This study investigated the phenolic compounds and antioxidant capacity of fruit extracts of Opuntia dillenii (Ker Gawl.) Haw.(OD) and Opuntia ficus-indica (L.) Mill.(OFI), yellow (F1) and red (F2) varieties. In order to evaluate the antioxidant activity of these extracts on human sperm quality after thawing, the semen parameters (vitality, motility, acrosome reaction, oxidative stress, and DNA fragmentation) were analysed after 1 h of exposure. The results showed that OD has higher phenolic content and antioxidant power than OFI, and that they are higher in F2 than F1. Furthermore, regarding the activity of extracts on thawed sperm, the results showed a significant increase in motility in samples treated with OFI F1 and OD extracts, while an improvement in vitality and acrosome reaction and a reduction of DNA fragmentation were observed in all exposed samples compared to the control. Finally, a reduction of oxidative stress was observed in samples exposed to OFI F2 and OD than control.

The nutraceutical value of pomegranate in the treatment of many diseases is well-documented and is linked to its richness in phenolic compounds. This study aims to evaluate the nutraceutical and genetic diversity of novel pomegranate genotypes (G1-G5) in comparison to leading commercial pomegranate varieties, i.e., 'Wonderful', 'Primosole', 'Dente di Cavallo' and 'Valenciana'. Morphometric measurements were carried out on fruits, accompanied by chemical characterization (total phenolic content, antioxidant activity, carbohydrates and minerals) and the development of four new polymorphic SSR markers involved in the flavonoid pathway. The cultivars displayed a marked variability in the weight and shape of the fruits, as well as in the weight of the arils and juice yield. The highest level of total phenolic content and antioxidant activity was found in 'Wonderful' and G4, while the lowest was in 'Dente di Cavallo'. Furthermore, the results showed that pomegranate juice is an excellent source of minerals, especially potassium, which plays a key role in organ functioning. The new flavonoid-related markers effectively differentiated the cultivars with the same diversity pattern as morpho-chemical characterization, so the SSRs developed in the present study can be used as a rapid tool for the identification of pomegranate cultivars with relevant nutraceutical traits, such as the new genotypes investigated.

The cardoon seeds presscake is a natural by-product resulting from the mechanical treatment of the cardoon plant seeds. The chemical composition suggests its possible use in animal nutrition due to the content of proteins, fibre, lipids and biologically active substances. The presscake studied contained 7.1% of fat, 26.8% of fibre, 23.9% of protein, 37.1% of nitrogen-free extract and a goodly phenolic content (13.36 mg GAE g-1 DW). The major fatty acids were oleic (26.5%) and linoleic (58.4%), showing a high degree of unsaturation. This study evaluated for the first time the inclusion of the cardoon presscake in pigs' diet. The effects of the dietary treatment on meat quality and fatty acids profile were assessed. The results indicate that the intramuscular fat (IMF) was higher in the untreated animals. The monounsaturated fatty acids (MUFAs) were lower in the IMF of the treated pigs, whereas the polyunsaturated fatty acids (PUFAs) were higher.

Cynara cardunculus L. is a multipurpose crop, characterized by high production of biomass suitable for energy purposes and green chemistry. Taking advantage of its already demonstrated ability to grow in polluted environments that characterize many world marginal lands, the aim of this work was to investigate the response of different cardoon genotypes to exposure to cadmium (Cd) and arsenic (As) pollution, in order to use this crop for rehabilitation of contaminated sites and its biomass for energy production. In this study, seeds of two wild cardoon accessions harvested in rural and industrial Sicilian areas and of a selected line of domestic cardoon were used, and the grown plants were spiked with As and Cd, alone or in combination, at two different concentrations (500 and 2000 µM) and monitored for 45 days. The growth parameters showed that all the plants survived until the end of experiment, with growth stimulation in the presence of low concentrations of As and Cd, relative to metal-free controls. Biomass production was mostly allocated in the roots in As treatment and in the shoots in Cd treatment. Cd EXAFS analysis showed that tolerance to high concentrations of both metals was likely linked to complexation of Cd with oxygen-containing ligands, possibly organic acids, in both root and leaf biomass with differences in behaviour among genotypes. Under As+Cd contamination, the ability of the plants to translocate As to aboveground system increased also showing that, for both metal(loid)s, there were significant differences between genotypes studied. Moreover, the results showed that Cynara cardunculus var. sylvestris collected in an industrial area is the genotype that, among those studied, had the best phytoextraction capability for each metal(loid).

This study compared the capacity of the seagrass Cymodocea nodosa and the green alga Ulva lactuca to sequester the trace elements Cd, Cr, Cu, Ni, Pb and Zn, in seawater and benthic sediments, under the same environmental conditions. Samples of C. nodosa leaves and U. lactuca thalli were collected in four sites off the coast of Sicily (Italy), with varying degrees of anthropogenic impact. C. nodosa accumulated generally higher levels of trace elements, suggesting a better performance in marine phytoremediation. In turn, C. nodosa and U. lactuca showed a similar performance regarding the bioindication of trace elements. Since salinity, temperature and pH had relatively constant values, their role in the accumulation processes in both species should be considered as secondary. Although C. nodosa and U. lactuca fulfill several criteria as bioindicators, including widespread distribution, abundance and tolerance to toxicity, their regular use in biomonitoring programs is still limited.

Cardoon (Cynara cardunculus L.) is a Mediterranean crop, member of the Asteraceae family, characterized by high production of biomass and secondary metabolites and by a good adaptation to climate change, usable in green chemistry, nutraceutical, and pharmaceutical sectors. Recent studies demonstrated the ability of cardoon to grow up in a stressful environment, which is associated with enhanced biosynthesis of biologically active compounds in these plants, and this effect is increased by abiotic stresses (salt, heat, pollution, and drought stress) that characterize many world marginal areas, affected by the climate changes. The plant response to these stresses consists in implementing different processes that modify some plant biological functions, such as alleviating both cellular hyperosmolarity and ion disequilibrium or synthesizing antioxidant molecules. The aim of this work was to investigate different cardoon response mechanisms to abiotic stresses and to evaluate their influence on the biologically active compounds biosynthesis. Following this purpose, we analyzed the ability of cardoon seeds to germinate under different salt stress conditions, and on the sprouts obtained, we measured the total phenol content and the antioxidant activity. Moreover, the growth of cardoon seedlings grown under heavy metals stress conditions was monitored, and the expression levels of heavy metal transport-associated genes were analyzed. The results showed the ability of cardoon plants to tolerate abiotic stress, thanks to different defense mechanisms and the possibility to obtain biomass with high content of biologically active molecules by exploiting the natural tolerance of this species for abiotic stresses. Moreover, we identified some important genes encoding for metal transportation that may be involved in arsenic and cadmium uptake and translocation in C. cardunculus. Then, this species can be considered as a promising crop for green chemistry and energy in marginal lands.

Accumulation and bioindication of trace elements were compared in three seagrasses growing in the Mediterranean Sea: Posidonia oceanica, Cymodocea nodosa and Halophila stipulacea. The levels of the elements As, Cd, Cr, Cu, Hg, Mn, Ni, Pb and Zn were investigated in water, sediments, and roots, rhizomes and leaves of seagrasses. Results showed that seagrasses can accumulate comparable levels of trace elements, but P. oceanica and C. nodosa showed higher mean values of element accumulation. Moreover, P. oceanica and C. nodosa may accumulate high element concentrations in their leaves, whereas in H. stipulacea restricted with the bulk of trace elements in roots and rhizomes. Seagrasses reflected to a different degree the levels of several trace elements in sediments, especially P. oceanica and C. nodosa, whose use as bioindicators is recommended. The future step for an effective use of seagrasses as bioindicators of marine pollution is to set up biomonitoring networks on a large scale.

This study aimed to shed further light on the capacity of the seagrass Halophila stipulacea to accumulate and biomonitor the elements As, Cd, Cr, Cu, Hg, Mn, Ni, Pb, and Zn, present in water and sediments. Results showed that the organs of H. stipulacea accumulate different levels of trace elements, whose concentrations decrease mainly in the order of roots>rhizomes>leaves. The seagrass H. stipulacea showed levels of trace elements similar to those found in other Mediterranean seagrasses, e.g. Posidonia oceanica and Cymodocea nodosa. This study showed that H. stipulacea could act as a promising bioindicator of several elements, present in sediments, including As, Cd, Cu, Mn, Ni and Zn.

Honey is a valuable food produced by bees from sugary substances that they gather in nature. The transformation the nectar into honey, by bees, is long and complex. Except for honey, where heavy metals are absent or are found only in traces, the bees and their products have always been considered excellent biomarkers of such contaminants. We have assumed that the absence of heavy metals in honey is due to the presence of a detoxification system in the digestive system of bees, which involves metallothioneins, proteins that have a role in the homeostatic control of essential and non-essential metals. We have placed the beehives in three different zones: industrial, urban and rural. Investigations were carried out with ICP-MS method for the detection of heavy metals in the guts of honey bees and honey. The metallothioneins have been identified by Immunohistochemical and Western-blotting analisys. The investigations have shown the presence of heavy metals only in bees guts but not in honey, while the presence of metallothionein has been highlighted only in epithelium of the honey sac, demonstrating the existence of an efficient system of detoxification of heavy metals.

The use of inulin in pasta improves the nutritional value decreasing the glycemic index in the blood after pasta ingestion but it compromises the sensory characteristics in terms of elasticity, firmness, bulkiness and adhesiveness. Thus, in this work, the impact of substituting whole-meal durum wheat flour with inulin on cooking quality, sensory and textural properties, digested starch and chemical composition of spaghetti was investigated. Two types of inulin at two different concentrations (2% and 4%) were adopted: inulin extracted from cardoon roots (CRI) (high polymerization degree) and commercial inulin (CHI) (low polymerization degree) produced from chicory. From the chemical point of view, the sample with 4% CRI showed the greatest total dietary fibres content and the lowest available carbohydrates content. A global acceptable quality was also recorded in all the other technological and sensory properties of enriched pasta with both types and both concentrations of inulin. The most feature of the work is that when CRI was added to the dough, better results were recorded, thus suggesting that for pasta enrichment, the selection of inulin with proper polymerization degree is a strategic factor for final product acceptance.

The suppression of the immune system creates a permissive environment for development and progression of cancer. One population of immunosuppressive cells that have become the focus of intense study is myeloid derived suppressor cells (MDSCs), immature myeloid cells able to induce immune-escape, angiogenesis, and tumor progression. Two different subpopulations have been identified and studied: granulocytic and monocytic MDSCs, with a different immunophenotype and immunosuppressive properties. Recently, an accumulation of both Gr-MDSCs and Mo-MDSCs cells has been found in the peripheral blood of chronic myeloid leukemia (CML) patients. They are part of the tumor clone showing BCR/ABL expression. Imatinib therapy decreases both MDSCs and arginase 1 levels to normal ones. This review will focus on actual knowledge for human MDSCs and their immunosuppressive activity in CML patients, with a critical attention to comparison of Gr-MDSCs and polymorphonuclear cells (PMNs). We will then suggest the monitoring of MDSCs in patients who have discontinued tyrosine kinase inhibitors (TKIs) therapy to evaluate if their increase could correlate with disease relapse.
